This is a question more out of curiosity rather than need, but when do you know it's time to repack a muffler? Will it sound different or shoot black smoke or something?

It will sound different or shoot black smoke IF you try to repack it!

Well...I believe Two Brothers recommends repacking after 3000 miles. However, I close to 4000 miles on mine and there has been no change in sound or performance. No smoke either.
